Goals from Marvin Johnson, Joe Rothwell and Conor McAleny fired the U's into a convincing first-half lead before Walker joined the action in place of Jermaine Pennant at the interval.
Having seen McAleny score his second shortly after the restart, Walker made an immediate impact as James Vaughan headed home his corner to pull one back for Bury just before the hour-mark.
However, there was to be no late comeback for the Shakers as McAleny completed his hat-trick in stoppage-time to wrap up the scoring for Oxford.
